If I had suspected that there was malware on a USB, will autoplays being disabled be enough for it to not affect me?

Also what sort of options can I enable on ESET that will reduce the likelihood of infection? Thanks.

Disabling the autoplay feature of Windows will not prevent potential autorun malware from being run manually. ESET detects any kind of threats, including autorun malware. However, there is no AV solution that would protect you from malware 100% with zero false positives so safe computing and keeping the system up to date is a must.
